Eduardo Salvio (89) struck late on to give Atlético a win in Madrid. Falcao (9) gave them an early lead with the club's 400th goal in UEFA club competition, which was cancelled out by Mame Diouf (38).
Hannover's only previous contest with Spanish opponents came in this season's UEFA Europa League play-off round when they eliminated Sevilla FC, winning 2-1 at home then drawing 1-1 away.
Die Roten have yet to lose a home game in UEFA competition, recording five wins and two draws over the course of their two continental campaigns. Atlético, meanwhile, have lost just one of their last eight European away games.
The Madrid club last made the quarter-finals in 2010 and went on to lift the trophy. They must improve their record in Germany (W4 D3 L7) if they are to do the same this year.
